Get back to the basics first... it usually isn't something complicated. Remember, fuel, spark, air, compression. All in the right amounts at the right time. Here's a few humble suggestions...
Are you sure you are actually on TDC? Did the timing chain skip a tooth on you when you were trying to get the sprocket to go onto the end of the cam?
Are you sure the distributor isn't one tooth off on the cam gear? It easy to do, and will chug along at about 800rpm full throttle.
Are the plug wires all run correctly? You don't have 2 and 3 swapped or anything?
All of these have happened either to me or to someone else when I was there. Something real small can seem really complicated and make you want to light the whole damn thing on fire... trust me. Good luck.
